<i>Neisseria gonorrhoeae</i> Ketol-Acid Reductoisomerase Is a Potential Therapeutic Target

The host-adapted human pathogen Neisseria gonorrhoeae is the causative agent of sexually transmitted infection gonorrhea. increased emergence gonorrhea infections worldwide, associated with surging resistance to antimicrobial treatments alarming. Antimicrobial (AMR) a global threat health and occur through various molecular mechanisms. This research aims identify therapeutic targets in N. gonorhoeae as potential antibiotic adjuvant. work focused on ketol acid reductor-isomerase enzyme (KARI), an involved branched-chain amino acids biosynthesis. A BLASTp analysis revealed that KARI highly conserved strains present important bacterial pathogens including ESKAPE. Sequence alignment different proteins from gut microbiota demonstrate residues forming active site cofactors binding sites are among all tested KARIs. 3D homology-based model for gonococcal was generated using Swiss server template S. aureus. shows this adapts conformation upon cofactors, allowing substrate catalysis, while closed state.
